ATCC 750 热带念珠菌使用说明
组成：冻干菌种瓶、菌种复溶液（用于复溶菌种及保存用）、使用说明书。
方法：建议用下列方法恢复培养冷冻干燥的菌种。
复苏菌种前应准备适于该菌种生长的固体或液体培养基和相应的培养设备，所有操作均应在符合生物安全保护及无菌条件下进行。
－、疫苗瓶开封
1．拉开易撕铝盖，用浸过75％酒精的脱脂棉擦净丁基胶塞及疫苗瓶。
二、菌株恢复培养
1．吸取适量的菌种复溶液，滴人疫苗瓶内，轻轻振荡，使冻于菌体溶解呈悬浮状。
2．取适量菌体悬浮液(建议岢养菌种可适当加大用量)，移植于沙保罗琼脂平板上，放置在30-35℃培养24-48小时.真菌在23-28℃培养箱中培养24-72小时（必要时可延长培养时间）
3．斜面（一代）放于4℃，并用封口膜密封平板，每半月转接一次，代数一次类推，建议使用菌种代数不超过5代，5代后菌种应销毁。
三、注意事项
1． 菌种活化前，请保存在2-8℃的环境下。
2． 嗜血杆菌、奈瑟氏菌属等初代培养需要5-10%CO2环境和的培养基，建议使用培养基。
3． 厌氧菌的培养，如无特别说明，自开封至接种完成，均需以无氧气体充填，以保持厌氧状态。
4． 某些菌种经过冷冻干燥保存后，延迟期较长，需连续两次继代培养才能正常生长后使用。
